Lysosomal acid lipase deficiency (LAL-D) is a rare genetic disorder with two clinical variants: Wolman disease in infants and cholesteryl ester storage disease in children and adults. Sebelipase alfa, a recombinant human LAL, has been evaluated for its safety and efficacy in treating LAL-D. A systematic review of seven records from five studies involving 110 patients showed significant improvements in serum transaminases and serum lipids, though changes in gamma-glutamyl transferase and liver volume were not significant. Adverse events were mostly mild-to-moderate. Sebelipase alfa is concluded to be an effective and well-tolerated treatment for both variants of LAL-D.
